Does the number change when you factor in boards ... and which number is more relavant?
I'm thinking of a situation like a board of 46TJ when someone has a hand like A4K9 where they can theoretically make a low as they have two low cards, however it is not possible in pratice as they pair one of their low cards on the board.
Of course I could just be misunderstanding things ...

Those numbers are as if the 4-card hand was dealt from the top of a full 52-card deck. Obviously once you start adding in dead cards you will affect that distributions to some degree. You can estimate how this might happen in-game by editing the sim and entering board cards; as you proposed http://goo.gl/7eXc
However, be aware that "actual" ranges will not necessarily discount in the same way. This is because people aren't generally playing 100% of hands preflop. One way to estimate this is to repeat the calculation but asking only for the hands that fall in the top 30% of hands, which gives a slightly different distribution.

(The sim breaks if you ask for "top 30% and quads", I reported this bug to the site, needles to say quads are not in the top 30% anyway; AAAA is ranked in the 32nd percentile).
