2nd/3rd barrel simply means continuing with your betting past the flop. It's generally used to refer to yourself bluffing or about villains.

bet flop = 1st barrel
bet turn = 2nd barrel
bet river = 3rd barrel

eg.

"Should I fire the 3rd barrel?" is asking whether to bluff the river after betting flop and turn.
"The guy 3barreled me on QK526" means the villain bet all 3 streets.

What should his range be for taking this c/r on turn in 3betpot line?
Does it change when you bet a more standard smaller ammount?
great video btw

There's no such thing as a 4barrel in hold'em... maybe in stud or something? Grin

Betsize is irrelevant, only that a bet happened matters.
